The ADP-glo (i.e. kinase activity) assay used to generate the validation and test sets is focused on active site inhibitors, i.e. compounds binding the active site of this kinase. DEL data are provided with and without a known active-site inhibitor. Focusing on compounds with significantly higher read counts in the absence than in the presence of this small molecule inhibitor should lead to an enrichment in molecules binding at the active site of PGK2, i.e. the type of compounds in the validation and test sets. We strongly recommend that participants account for this in their hit selection strategy.
